The vascular bone marrow niche is a complex system, crucial for the production of erythrocytes, platelets and immune cells derived from hematopoietic stem progenitor cells (HSPCs). It supports normal homeostasis but up-regulates hematopoiesis after injury to stimulate recovery, a process that is regulated by numerous molecular and cellular mechanisms and is still not fully understood. There is however strong evidence that extracellular matrix (ECM) and mesenchymal stem cells (MSCs) in the vascular bone marrow niche are important for a normal function of HSCs. Therefore we hypothesize that physical forces and specific adhesive signals derived from the bone marrow extracellular matrix (ECM) control functions of bone marrow-specific endothelial cells, MSCs and HSCs that are key to hematopoiesis. The aim of this study is to investigate the effect of ECM on different cell-ECM and cell-cell interactions within the vascular bone marrow niche with as an ultimate goal mimicking the human bone marrow niche in an in vitro 3D model to culture and expand MSCs and HSPCs for transplantation purposes.
