Abstract
Hematopoietic stem and progenitor cells (HSPCs) are the core component of hematopoietic cell transplantation (HCT), a last-resort curative therapy for >50.000 patients annually suffering from hematologic, oncologic, and immunologic diseases. Despite its widespread clinical use, the mechanisms by which transplanted HSPCs regenerate blood (or fail to do so), and how transplantation shapes the quality and clonal composition of the recipient’s stem cell pool remains incompletely understood. Recent advances in single-cell genomics have transformed our ability to study HSPC mutagenesis, differentiation trajectories and tracking of clonal dynamics during steady-state hematopoiesis, regeneration, and disease. In this seminar, I will present our recent work into the origins and clonal dynamics of hematopoiesis after transplantation and discuss their implications for donor selection and long-term surveillance of HCT recipients.
