'New development in the preparation and storage of platelet concentrates from buffy coats'

Research interests
Proficiency testing Yearly organization of 5 national proficiency studies: Platelet counts in platelet concentrates; leucocyte counts in blood products; pH, blood gas, and metabolite measurements in platelet concentrates; CD62p and Annexin V expression in platelet concentrates; trombocyte count using CD41 in platelets concentrate. Product development Cryoseal (in Dutch called cryolijm) is a fibrin glue of single donor plasma which Sanquin has started to produce as an alternative for autologous or pooled plasma fibrin glues. Studies have been performed to elongate the storage time. Furthermore validation studies are ongoing to produce cryoseal from whole blood derived plasma and to produce cryoseal with a longer clotting time to broaden the use of cryoseal. Cryo-poor plasma, a by product of cryoseal production, might be a possible therapy for patients with TTP. A Hemotopoietic Progenitor Cell (HPC)-simulant has been developed which can serve as a substitute for patient’s material to be used for validation studies. Relation between in vivo and in vitro parameters During a period of 1.5 years all platelet concentrates were prepared in a BCSI container. This storage bag enables pH measurement at any time during storage without sampling, i.e. in a sterile way. The pH of the platelets concentrates was studied to determine whether or not there is a correlation with the in vitro quality and/or bacterial contamination, storage time and the yield of the transfusion in patients.
