Lymphocyte cytokine profiling (ICS)
Monitoring cell populations on the basis of phenotypic markers can provide useful information about immune reactions in patients (Cellular Immunophenotyping (flow cytometry)). Additionally, much more information can be obtained by investigating the functionality of specific immune cells. T cells produce cytokines and/or granzymes upon stimulation with their antigen. The percentage of T cells and the type of cytokines that are produced give information whether a T cell responds in a suppressive or stimulatory manner. We can distinguish between Th1, Th2, Th17, Tfh and T regulatory cytokines responses. Especially when combined with antigen specificity (CD40L assay and CD8: MHC multimer analyses (combinatorial coding)), this type of assay can help unravel the polarization, strength and specificity of immune responses to immune modulating therapies.
We have a broad range of cytokines that can be measured simultaneously, thereby reducing the amount of cells needed to perform these types of assays.
PBMCs from a healthy donor were stimulated with PMA/Ionomycine for 4 hours. The cells were gated on CD3+ and CD4+ cells, showing IL-2 and IL-4 production.
